臺灣原生花臉香蘑栽培特性及其生物活性之探討

花臉香蘑(Lepista sordida (Fr.) Singer)是一紫色及具有薄荷清香的野生食用菇種,因不易栽培至今仍無法規模化量產。本研究發現臺灣本土花臉香蘑菌株以24-28 ℃菌絲生長最為良好,CEA為其最佳的培養基。以洋菇高產室內發酵堆肥大量栽培花臉香蘑LS-2菌株,覆土後28.3日可開始採收,產量達2.6 kg/筐(15 kg堆肥)、生物效率43.5%。LS-2菌株子實體總多糖含量13.41%、總多酚含量10.66 mg/g、總花青素含量0.49 mg/g。本研究評估以臺灣本土獲得的花臉香蘑菌株培育而成的子實體之水粗萃物(WE-LS)及乙醇粗萃物(EE-LS)對樹突狀細胞之影響。

證實花臉香蘑水粗萃物經由TLR-2和TLR-4訊號傳遞途徑促進樹突狀細胞之成熟及活化,進而提升樹突狀細胞誘導Ag-specific Th1細胞反應之能力。以對樹突狀細胞幾乎沒有毒性的EE-LS劑量來評估其對LPS誘導樹突狀細胞成熟及活化之影響。實驗證實EE-LS以劑量依賴性方式顯著降低LPS誘導的CD40、CD80和CD86表現量及同種異體T細胞增殖的能力,EE-LS亦可抑制LPS引起樹突狀細胞之與發炎反應相關的細胞激素(如TNF-α、IL-6及 IL-12等)及NO/iNOS mRNA之表現量,也進一步證實EE-LS對LPS誘導樹突狀細胞活化及促發炎因子分泌之抑制作用是經由抑制LPS誘導NF

-κB活化的訊號傳遞途徑,而不是MAPK及Akt訊號傳遞途徑。本研究亦發現EE-LS對A549及CL1-5肺癌細胞株有毒殺作用,尤其對於高轉移性的肺癌細胞CL1-5有更顯著之細胞毒殺作用。EE-LS引起A549及CL1-5肺癌細胞死亡是經由調控細胞凋亡相關蛋白,如Bax、Bcl-2、XIAP及Survivin蛋白的表現,進而降低粒線體膜電位並釋放cytochrome c至細胞質引起caspase-9及其下游之caspase-3的活化之內源性細胞凋亡途徑導致肺癌細胞株細胞凋亡。本實驗也以異種移植腫瘤小鼠模式證實EE-LS在小鼠體內具有抑制腫瘤生長效果。因此,由本研究得知花臉香蘑菌株子實體粗萃物中

應該含有相當有效的免疫調節、抗發炎與抗癌之成分,然而這些活性成分有待未來再進一步的研究確認。
