魚類的鰓是直接與環境水域接觸的器官,俱有許多功能,包含呼吸和滲透壓調節。因此,鰓上的細胞需要持續地維持其功能,包括細胞本身的能量運用、滲透壓平衡、甚至新舊細胞的更新,以便適時反應外在水域環境變化。水域環境的變化包含其溶質的濃度變化和影響溶質溶解的因素--溫度的變化;淡水與海水環境中的鹽度差異即是前者。廣鹽性魚類具備可生存在淡水和海水的能力,其鰓表皮細胞也因應各種環境鹽度而有不同的滲透壓調節功能;因此,溫度可能影響廣鹽性魚類的鰓,造成其不同的生理反應。恆河稻田魚 (Oryzias dancena) 由於來源穩定、馴養方便、鹽度和溫度適應性皆良好,本論文以其為實驗模式魚種,以次世代定序分析獲得轉

錄體基因資料資料庫和蛋白質表現測定等方法,來比較在淡水和海水馴養的恆河稻田魚在面臨兩種不同程度的低溫 (降低攝氏10°C之18°C與最低耐受溫度15°C) 時,鰓的能量運用、滲透壓平衡、甚至新舊細胞的更新的變化,以及與粒線體生源變化之相關影響。利用次世代定序的結果,本論文研究首先進行轉錄體序列的重新組裝 (de novo assembly) 並以擁有最佳N50序列長度之結果完成重新組裝,接著進行GO和KEGG分類與命名,最後將命名之序列作為模板,並分析其在不同鹽度及溫度環境中的表現量。第二部分實驗結果發現,馴養在淡、海水的個體,低溫也對其上鰓的離子調節功能有不同的影響,淡水個體受到15°C低溫

本論文分成兩大部分,一為仿刺參與九孔、白蝦混養,評估台灣的冬天是否適合進行混養與探討混養對成長的影響,二為人工飼料的開發:以魚粉與發酵豆粉作為蛋白質來源,探討最適動植物蛋白質比例與最適蛋白質含量。實驗一為建立仿刺參與九孔混養的養殖技術,十二月至隔年五月以初重29 g之仿刺參於台灣北部與九孔混養,結果顯示養殖九十天後即可達上市體型,陸地池的特定成長率(specific growth rate)為1.59 ± 0.02 %,高於海底池的特定成長率1.57 ± 0.01 %(p < 0.05)。實驗二於室外流加水系統中以密度為60 ind/m2的白蝦與不同密度(21、42 ind/m2)的仿刺參混
