乙醯輔酶A (Acetyl-CoA)是所有生物代謝途經中的核心代謝產物,可被用於檸檬酸循環氧化產生能量並提供電子給其他生化反應使用,亦作為包括:脂肪酸、類萜 (Terpenoids)、聚酮 (Polyketide)等合成途徑在內的重要前驅物,利用乙醯輔酶A設計並開發之代謝途徑因而具有廣泛的應用性且可被建立於不同物種中。雖然乙醯輔酶A相關代謝途徑具有很高的發展潛力,然而,若想轉換乙醯輔酶A回到上游代謝物丙酮酸的天然代謝途徑幾乎都需要兩個乙醯輔酶A經脫碳途徑產出一個丙酮酸而減少產率,無碳損途徑如丙酮酸合酶 (Pyruvate synthase) 則具有低活性與厭氧環境之需求,因此,開發一個無碳損

乙醯輔酶A至丙酮酸的代謝途徑具有很大的價值。本研究中基於乙醯輔酶A的衍伸代謝途徑的發展潛力與面對的困難,首先於大腸桿菌中建立以乙醯輔酶為前驅物之丁醛與乙酸丁酯合成途徑,藉由抑制競爭基因、代謝途徑設計、酵素表現量優化、產物移除……等代謝工程策略,達到高產量之生質化學品生產。接著,有鑑於部分物種(如藍綠菌)乙醯輔酶A濃度低難以推動下游代謝物合成,我們建立了一個代謝途徑迴路 (Balanced ATP driving force module) 以提供反應途徑驅動力,並於藍綠菌中證明其能有效提升由乙醯輔酶A合成3-羥基丁酸。最後,本研究建立一經3-羥基丙酸之合成代謝途徑,提供無碳損代謝途徑達成乙醯輔

酶A至丙酮酸之轉換,此途徑可被應用於提升丙酮酸衍生產物產率、二氧化碳再利用等重要代謝工程問題。綜上所述,本論文以乙醯輔酶A的泛用性為基礎,開發多種代謝工程策略以建立高效率乙醯輔酶A轉換平台。

組織工程,又稱為再生醫學,是現代醫學中十分重要的一部分,促進傷口附近的血管重生,進而達到組織或是器官修復正是再生醫學所追求的目標。而一個理想的組織工程需要的細胞支架必須能讓細胞貼附生長並整合血管再生,加速受傷部位的恢復速度。在這項研究中,聚(癸二酸甘油酯)(PGS)是一種具有生物相容性,可以運用在血管組織工程修復上的材料。研究發現,PGS是具有生物相容性的彈性體,在體內、外都能夠進行降解,是可以運用在組織工程中理想的生物材料之一。在本研究中,PGS由癸二酸和甘油以等莫耳比聚合而成,並加入由羥基聚(3-羥基丁酸酯) (PHB)醇解酯化反應成的雙羥基聚(3-羥基丁酸酯)( PHB-diol)來合

成複合材料支架,PHB-diol以0.05、0.1以及0.15的莫耳比例取代部分原先欲與癸二酸聚合而成的甘油,進行聚合反應以後,可以得到不同比例的共聚彈性體,由於PHB-diol本身脆硬與其在降解時副產物能夠協助細胞培養的特性,我們期待加入PHB-diol之後不僅能助於細胞生長,同時改善PGS的機械性質。此外,本研究亦結合了先前實驗室以氯化鈉作為製孔劑在材料表面創造出微孔洞來讓支架提供更適合細胞生長的環境,藉以發展更適合細胞培養之生物材料。本研究中顯示,加入PHB-diol的比例越高,材料的交聯度越低,這是因為PHB-diol分子與甘油分子分子量差異巨大所導致。而加入PHB-diol可以提升P

GS材料本身的機械強度,由拉伸試驗的結果得知,加入PHB-diol的量越多,材料能夠承受的最大應力也隨之提升,能加強PGS機械強度不足的弱點。考慮到可能植入生物體內,材料的親疏水性也是必須考慮的,PHB的加入使複合材料更加的親水,但後續交聯程度降低時,也影響到親疏水性,使材料總體性質並不單純隨著PHB量增長而改變。以掃描式電子顯微鏡觀察材料表面的微結構,在加入作為製孔劑的氯化鈉顆粒前,材料表面平整,但加入氯化鈉顆粒並清洗去除乾淨後,可以看到均勻分布的微孔洞。透過細胞培養在材料上的結果顯示,PGS-PHB-diol複合材料並沒有明顯的細胞毒性,L929小鼠神經纖維母細胞跟內皮前驅細胞(EPCs)

的WST-1結果皆是如此。整體來說, PGS是一種可以讓細胞貼附生長的生物可降解材料,具有能提供細胞生長的環境條件,在血管組織工程上存在廣泛應用的潛能。
