以神經網路進行灰階影像上色任務容易有輸出顏色黯淡之問題,有論文提出此來自於自然影像中顏色機率偏向低彩度之現象。而色彩空間為上色任務中鮮少被研究之主題,我們研究是否該現象於各種色彩空間皆會發生,並發現統計自然影像顏色機率於以基於色環(Hue)與飽和度(Saturation)設計之色彩空間時,該現象之嚴重度遠比在上色任務中常用的色彩空間Lab上統計時來得小。我們以色彩空間定義出發,推論出除了自然影像之統計特性外,此現象之根本原因來自於RGB色彩空間映射到一色彩空間後於各色度上之投影量的差距。 在上述機率方面表現較好的色彩空間中,我們探討他們於上色任務的適合性,推論出其中最適合的色彩空間為H

SL,並針對HSL中HS值之不穩定性以色彩差異的角度推論此現象已被亮度的機率分布自行解決。接著我們基於此色彩空間設計一套簡易而有效的演算法,實驗證明HSL在以像素級損失訓練之神經網路上,在色彩鮮豔度上皆優於Lab。我們也將引用數量相當高的兩篇上色任務論文中的Lab色彩空間調換成HSL,並證明在多種評估指標上皆有顯著進步。最後我們分析HSL在以非像素級損失訓練之神經網路上會碰到的隱憂,供後續研究者參考。

This doctoral dissertation focuses on two parts; the first part demonstrates the polarization-resolved (PR) stimulated emission pump-probe microscopy, and the second part represents the implementation of photon-counting based stimulated emission pump-probe microscopy. We have realized the polarizat

ion-resolved fluorescence lifetime measurements through stimulated emission pump-probe (stimulated gain (SG) and stimulated spontaneous loss (SSL)). SG is applied to realize the polarization-resolved fluorescence lifetime measurement through stimulated emission, which promises much higher temporal

resolution (~4 ps) than conventional time-correlated single-photon counting (TCSPC). The rotational correlation time (RCT) and polarization anisotropy (PA) of ATTO 647N dye are characterized via various temperatures. The results reveal the need for high temporal resolution via hetero Förster energy

transfer (Hetero-FRET) through the interaction between the gold nanorods (GNRs) and the fluorescent dye ATTO 647N. Notably, our results highly favourable with the conventional TCSPC method, which is rather limited in temporal resolution, for the above characterization. Additionally, this technique i

s applicable even under ambient light while being very cost-effective and robust.Furthermore, SSL demonstrates the contrast enhancement through polarization-resolved pump-probe microscopy, implemented by measuring the spontaneous fluorescence loss induced by stimulated emission. The pump-probe lifet

ime measurement technique has been compared with conventional fluorescence polarization microscopy. The anisotropy values thus obtained are 0.11 and 0.01, respectively. The contrast improvement is attributed to the multiphoton mechanism in sharpening the point-spread function and the polarization re

Also, the pump-probe technique promises higher temporal resolution in lifetime measurements than TCSPC, enabling more precise determination of the fluorescent molecules' rotational diffusion time constant, often in the sub-nanosecond regime.To overcome the limitation of analogue measurement

s in SSL, photon-counting based stimulated emission pump-probe microscopy is employed through time-correlated single-photon counting (TCSPC). The experimental measured stimulated photons quenching efficiency is compared with theoretical photon statistics. The accurate fluorescence lifetime measureme

nts of ATTO 647N indicates the effectiveness of this method. Notably, atomic calculations are used to obtain absolute cross-sections.
