抗生素的濫用導致全球抗藥性問題嚴重，為緩解其帶來的傷害，科學家積極尋找低抗藥性物質。其中，乳鐵蛋白素 (lactoferricin, Lfcin) 為乳鐵蛋白胃蛋白酶水解物所分離出的抗菌胜肽 (antimicrobial peptides)，對細菌和真菌具有廣效抗菌活性且有不易產生抗藥性的優點，於減緩抗藥性問題上頗具潛力。然而，產製 Lfcin 成本高且耗時，故本研究乃嘗試透過基因轉殖技術以嗜甲醇酵母菌 (Pichia pastoris) 蛋白質表現系統產製 Lfcin，並尋求較佳的抗菌活性。本研究以 pPICZA 質體構築包含 α 因子訊息肽 (α-factor signal pe

ptide, α-SP) 的外泌型載體和無 α-SP 的胞內型載體，表達 Lfcin 一倍體或二倍體 (Lfcin2) 基因序列：pPICZαA-Lfcin、pPICZA-Lfcin、pPICZαA-Lfcin2 及 pPICZA- Lfcin2，轉染 P. pastoris 並比較其表現 Lfcin 之抗菌活性、產量等優劣性。結果顯示，以 0.5 % 甲醇誘導 48 小時後，胞內型轉殖株 pPICZA-Lfcin/P. pastoris 和 pPICZA- Lfcin2/P. pastoris 表現之重組 Lfcin 較外泌型轉殖株 pPICZαA-Lfcin/P. pastoris 和 p
