全固態電池現今是個極具發展性及有趣性的研究領域,能避免大量液態電解液造成潛在的爆炸、漏液危險,且能直接使用鋰金屬當作負極,透過減少體積來提高能量密度,而電解質中又以固態硫化物電解質最為突出,因其擁有最高的導離子度與熱穩定性。但組裝出硫化物全固態電池需要在惰性氣氛下進行,並且要克服介面接觸不良以及副反應問題。本研究分為兩個部分,一為全固態電池的組裝,從錠狀電池到膜狀電池,並探討正極、負極、固態電解質的各個參數的影響。使用LNO@NCM811高鎳三元材料當作正極,Li6PS5Cl作為固態電解質,鋰與銦金屬作為負極,1 wt %的添加碳,第二部分為軟包電池組裝,成功組裝出3x3 cm2大小的NMC

背景  心室心律不整與發炎反應的活化以及心臟衰竭有很密切的關係。其中介白質-17可誘發發炎反應,是免疫發炎的早期物質,並且於心肌缺血以及梗塞後發炎反應中上升。介白質-17與誘發出心室心律不整的機制尚不清楚。先前研究顯示較陡之動作電位回歸曲線、較多心室顫動相位奇點(phase singularities)及發炎前驅物質會誘發心室心律不整。心臟衰竭為常見之心臟疾病,容易誘發出心室心律不整進而導致死亡。血管張力素受體-腦啡肽酶抑制劑為新一類治療心臟衰竭藥物,能有效降低心臟衰竭惡化造成的死亡、並減低猝死發生率。心臟衰竭大規模臨床試驗(PARADIGM-HF trial)表明,血管張力素受體-腦啡肽酶

抑制劑治療有效地減少了心因性死亡,可能透過抑制心室心律不整。目前文獻尚未有探討發炎前驅物質介白質-17以及血管張力素受體-腦啡肽酶抑製劑之間的相互關係,本論文目的在研究發炎前驅物誘發心室心律不整之機轉與以血管張力素受體-腦啡肽酶抑製劑治療之方向。方法   實驗一:將正常兔子心臟分離,懸吊於離體心臟灌流系統中。第一組為控制組,第二組為介白質-17組,第三組為介白質-17中和劑組;實驗二:利用左前降支中段完全結紮4週後建立心臟衰竭的兔子模型。實驗二,組別分別為,第一組為正常組,第二組為心臟衰竭組,第三組於結紮一周後連續給予三周血管收縮素受體阻斷劑,第四組於結紮一周後連續給予三周血管張力素受體-腦啡

肽酶抑製劑。利用心臟光學標測成像系統透過高速相機與軟體研究動作電位,測量左心室之動作電位時間、動作電位變異度、鈣離子瞬變電流時間、傳導速度、心室顫動相位奇點、鈣離子流的最大變異程度、動作電位恢復曲線及心室心律不整之誘發率。同時測定心電圖、發炎前驅物質、離子通道表現以及心室纖維化嚴重程度。結果  在正常心臟中加入介白質-17發現在不同的電刺激長度下,動作電位和鈣離子瞬變電流時間延長、鈣離子流的最大變異程度縮短、傳導速度變慢,並且增加了動作電位電氣交替以及鈣離子瞬變電流時間電氣交替閾值、動作電位恢復曲線的最大斜率導致容易觸發心室心律不整發生。在心室纖維顫動的引發時所形成的相位奇點以及主頻,介白質-

17組別顯著高於中和劑組別以及控制組。介白質-17組別於鈣離子相關通道中,發現NCX、PLB、RyR 通道表現量增加,並且於Cav1.2、SERCA2a通道表現量則是降低。介白質-17藉由增強動作電位間距電氣交替以及鈣離子間距電氣交替閾值調控鈣離子平衡,進而增加發生心室心律不整發生率。  心臟衰竭組與血管收縮素受體阻斷劑組和血管張力素受體-腦啡肽酶抑製劑組相比,心室更為肥厚且較低的射出分率以及縮短分率,動作電位延長、傳導速率較慢、有較長的QRS波、QT波和 QTc 間距。心臟衰竭組心室纖維顫動引發時所形成的相位奇點增加、心室心律不整誘導性以及動作電位恢復曲線的最大斜率均高於血管收縮素受體阻斷劑

組和血管張力素受體-腦啡肽酶抑製劑組。與心臟衰竭組相比,血管收縮素受體阻斷劑組和血管張力素受體-腦啡肽酶抑製劑組的纖維化程度較少、CD4+和CD8+細胞計數較低。結論  介白質-17藉由增強動作電位時間電氣交替以及鈣離子瞬變電流電氣交替閾值、影響調控鈣離子平衡,進而增加發生心室心律不整發生率。血管張力素受體-腦啡肽酶抑製劑可以透過逆轉電位持續時間、傳導速率、最大斜率、離子通道、發炎反應和纖維化進而有效抑制心臟衰竭造成的心室心律不整。藉由研究介白質-17對心室心律不整之影響,可找到治療的新方向。
